Chromogenic TAL Assay (Chromogenic endotoxin test assay)

TAL reagent is the lyophilized amebocyte lysate which is extracted from the blue blood of Limulus polyphemus or Tachypleus tridentatus.

Endotoxins are amphiphilic lipopolysaccharides (LPS) located in the outer cell membrane of gram-negative bacteria. Parenteral products contaminated with pyrogens including LPS can lead to the development of fever, induction of inflammatory response, shock, organ failure and death in human. Therefore, countries around the world have formulated regulations, requiring that any drug product claiming to be sterile and non-pyrogenic should be tested prior to release. Gel-clot TAL assay was first developed for bacterial endotoxins test (i.e. BET). However, other more advanced methods of the TAL assay have emerged. And these methods will not only detect but also quantify the presence of endotoxins in a sample.

Apart from gel-clot technique, techniques for BET also contain the turbidimetric technique and the chromogenic technique.
